The AIRE gene (autoimmune regulator) has been identified as an important mediator of central tolerance against ectopically expressed peripheral antigens. Mutatations of AIRE are responsible for autoimmune polyendocrinopathy syndrome type I (APSI) in human characterized by a multiorgan autoimmune disease.
